Synthesis and Immunological Study on the Peptide Corresponding to Antigenic Domain of the Sperm-specific Lactate Dehydrogenase-C4






Abstract
Lactate dehydrogenase C4(CDH-C4) is an antigenic protein found only in mature spematozoa and testis. It is known that immunization with sperm-specific isozyme of LDH-C4 results in the production of highly specific antisera and the fertility
supression
of female animals. It would be useful to replace the native protein with synthetic peptide that carries an antigenic determinant. A small peptide bearing an antigenic domain was selected by considering the hydrophilicity, hydropathy, and
evolutionary
variability.
The selected peptide MC(308-319), which is in the region of the highest local hydrophilicity, was synthesized by solid phase peptide synthesis and was immunized with nitrocellulose-coated peptide, was tested sera by immunodotting and enzyme
linked
immunosorbent assay to know whether this could elicit antibody production. Results shoed that rabbits immunized with peptide MC(308-319) produced antibodies that bound specifically to the immunizing peptide.
